スルフォラファンは、試験管内において終末糖化産物(AGE)形成を抑制する

慢性的な高血糖状態では、還元糖による蛋白質や脂質のアミノ基の非酵素的糖化反応が進み、循環血液中や組織で終末糖化産物(AGE)が促進的に形成、蓄積されてくる。近年、AGEの蓄積が、糖尿病の臓器合併症にかかわっていることが明らかにされてきた。本研究では、スルフォラファンの試験管内におけるAGE形成阻害作用について検討した。スルフォラファンはいずれの濃度(0-100μM)においても0~3日目までのインキュベーションでは、AGEに特異的な蛍光波長の発生に影響を及ぼさなかった。しかしながら、5日間のインキュベーションでは、いずれのスルフォラファン濃度においてもAGEに特異的な蛍光強度が低下することが観察された。さらに、ELISA法により、グリセルアルデハイドに由来するAGEを特異的に計測してみると、高濃度(25,100μM)のスルフォラファンにより濃度依存性にAGEの形成が抑えられることが見い出された。スルフォラファンは、AGEの形成を抑えることで、糖尿病の臓器合併症の発症・進展に保護的に働く可能性が考えられる。

Reactive derivatives from non-enzymatic glucose-protein condensation reactions, as well as lipids and nucleic acids exposed to reducing sugars, form a heterogeneous group of irreversible adducts called “AGE (advanced glycation end product)”. The formation and accumulation of AGE have been known to progress at an accelerated rate under diabetic conditions. There is a growing body of evidence that AGE and their receptor (RAGE) interaction elicit oxidative stress generation and evoke inflammatory and thrombogenic reactions, thereby being implicated in diabetic vascular complications. In this study, we examined the effects of sulforaphane on AGE formation in vitro. 0.4-100 μM sulforaphane for 5 days significantly decreased the AGE-evoked fluorescence intensity (Ex=355nm, Em=460nm). Furthermore, enzyme-linked immunosorbent assay revealed that sulforaphane at 25-100 μM dose-dependently reduced AGE formation in vitro. Our present study suggests that sulforaphane might protect against organ damage in diabetes via suppression of AGE formation.

ブロッコリースーパースプラウトの摂取は,血中AGEレベルを低下させる

【要約】

加齢に伴い生体内蛋白は糖化変性修飾を受け、構造的にも機能的にも劣化した終末糖化産物(advanced glycation end products:AGE)を形成するに至る。さらに、AGEは、細胞表面受容体であるRAGE(receptor for AGE)によって認識され、酸化ストレスや炎症反応を惹起させて、心血管合併症、アルツハイマー病、癌、骨粗鬆症などの老年疾患の発症・進展に関わることが知られている。われわれは以前に、試験管内において①スルフォラファンがAGEの形成を阻害すること、②スルフォラファンが血管細胞におけるRAGEの発現を抑えることを明らかにしてきた。本研究では、文書で説明と同意が得られた成人男女25名(男性11名、女性14名、平均年齢49.5歳)を対象に、スルフォラファン前駆体を高濃度に含むブロッコリースーパースプラウトを毎日25g、2ヵ月間摂取することで血中AGEレベルが低下するか否かについて、前向きに検討した。ブロッコリースーパースプラウトの摂取で、体重と体格指数(body mass index:BMI)が有意に減り、腹囲がおよそ2.5cm減少した。さらに、血中AGEレベルが24.5から19.6U/mLと顕著に下がり、HbA1c、拡張期血圧、総コレステロール、HDL-コレステロール、尿素窒素も有意に低下した。肝障害などの有害事象は、全く観察されなかった。また、体感と食事に対する意識に関するアンケート調査でも、被験者自身、体調の改善や減量を自覚しており、有意に食事に関する意識の向上が認められた。以上、スルフォラファン前駆体を高濃度に含むブロッコリースーパースプラウトを摂食することにより、生体内のAGE化が抑えられる可能性が考えられる。

Reducing sugars can react non-enzymatically with the amino groups of proteins to initiate a complex series of rearrangements and dehydrations, and then to produce a class of irreversibly cross-linked macroprotein molecules termed advanced glycation end products (AGE). The formation and accumulation of AGEs in numerous tissues have been known to progress in a normal aging process. There is a growing body of evidence that AGE and their receptor (RAGE) interaction evoke oxidative stress and inflammatory and reactions, thereby being involved in cardiovascular disease, Alzheimer’s disease, cancer growth and metastasis, and osteoporosis. We have previously found that sulforaphane not only inhibits the formation of AGE in vitro, but also reduces RAGE expression in vascular cells. In this study, we examined whether oral consumption of sulforaphane precursor-rich broccoli supersprouts could decrease serum AGE levels in humans. Daily intake of broccoli supersprouts for 2 months significantly decreased body weight, body mass index and waist circumference, and reduced HbA1c, AGE, diastolic blood pressure, total and high-density lipoprotein cholesterol, and blood urea nitrogen levels. It also raised consciousness of eating healthy food. Our present study suggests that intake of sulforaphane precursor-rich broccoli supersprouts could suppress the formation of AGE in humans.

ケルセチンの終末糖化産物(AGEs)形成阻害作用

【要約】

加齢に伴い生体内蛋白は一様に糖化を受け、臓器障害性の強い終末糖化産物(advanced glycation end products:AGEs)と呼ばれる劣化蛋白を形成するに至る。AGEsの生成、蓄積は、加齢や酸化ストレス、慢性炎症,高血糖下でも亢進し、老化のプロセスに深く関わることが明らかにされてきた。われわれは以前に、試験管内において25-100μMスルフォラファンがAGEsの形成を阻害することを報告した。そこで本研究では、低濃度ケルセチンのAGEs形成阻害作用について、スルフォラファンの共存、非共存下で検討を行った。0.4μMケルセチン存在下で3日間、0.4μMあるいは1.6μMケルセチン存在下で7日間、グリセルアルデハイドとウシ血清アルブミンをインキュベーションすると、AGEsに特徴的な蛍光活性が有意に低下することが見い出された。さらに、ELISA法により、グリセルアルデハイドに由来するAGEsを特異的に測定したところ、①0.4μMケルセチンによりAGEsの形成が有意に抑えられること、②0.4μMスルフォラファンを共存させることで、ケルセチンのAGEs阻害作用が強まることが観察された。ケルセチンは、AGEsの形成を抑えることで、老年疾患の発症・進展に保護的に働く可能性が考えられる。

A non-enzymatic reaction between reducing sugars and amino groups of proteins, lipids and nucleic acids contributes to the aging of macromolecules. Over a course of days to weeks, early glycation products undergo further reactions such as rearrangements and dehydration to become irreversibly cross-linked, macroprotein derivatives termed advanced glycation end products (AGEs). The formation and accumulation of AGEs have been known to progress at a normal physiological aging and at an accelerated rate under oxidative stress, inflammatory and hyperglycemic conditions. There is accumulating evidence that AGEs play a role in various aging-related disorders. We have previously shown that 25-100 μM sulforaphane inhibits in vitro-formation of AGEs. In this study, we examined the effects of lower concentration of quercetin on AGEs generation. When glyceraldehyde and bovine serum albumin were incubated with 0.4 μM quercetin for 3 days, 0.4 or 1.6 μM quercetin for 7 days, intensities of fluorescence characteristic to AGEs (Ex=355 nm, Em=460 nm) were significantly decreased. Furthermore, enzyme-linked immunosorbent assay revealed that 0.4 μM quercetin significantly prevented the formation of AGEs, and its effect was additive with 0.4 μM sulforaphane. Our present study suggests that quercetin might protect against aging-related disorders via suppression of AGEs formation.
